A multi-institution Chinese research team has tested topical clove oil, derived from Eugenia caryophyllata Thunb., as a treatment for canine sarcoptic mange, and published its findings in Frontiers in Veterinary Science [5]. The operational reason to care is not botanical: mange occurrence among unvaccinated shelter dogs and strays can top 26%, roughly one dog in four, while current chemical acaricides carry adverse effects and growing concerns about drug resistance and environmental sustainability [3][4][17].
The disease itself is a cost driver as much as a welfare problem. Sarcoptes scabiei mites infest the skin, the condition is contagious and zoonotic, and it can be difficult to detect [1]. Untreated, it progresses to bleeding sores, malnutrition, emaciation and secondary bacterial infections that can kill the animal [2]. Dogs of all ages, breeds and sexes are susceptible [3].
The trial design is straightforward. The team assembled 100 dogs with untreated mite infestations from breeders and shelters in Lanzhou, confirmed by microscopic examination of skin scrapings, drawn from about a dozen breeds of various sizes, and randomised them into five groups of 20 [9]. Group A received soybean oil alone as a negative control [10]. Groups B, C and D received 7% w/v clove oil in soybean oil at 0.05, 0.1 and 0.2 mL/cm respectively, a fourfold spread from lowest to highest dose [11][18]. Group E, the positive control, received 12.5% amitraz diluted in water at 0.05% per 0.1 mL/cm [12]. Soybean oil was chosen as the carrier for its established safety profile in veterinary topical applications, according to the paper [13]. Each group got one application every other day, four applications across a 14-day window, with symptom scoring for alopecia, erythema and exudation, pruritus and scaling, plus mite counts on days 0 and 14 [14]. Dogs left untreated were given ivermectin as full therapy once the study ended [14].
On mechanism, the researchers write that eugenol, clove oil's dominant bioactive component, binds NADH dehydrogenase chain 2 and inhibits mitochondrial respiratory chain complex I in the oxidative phosphorylation pathway, causing energy metabolism failure and mite death, and that compared with the classic mitochondrial inhibitor rotenone it shows better biosafety and highly selective toxicity against mite cells [7]. Acaricidal activity for clove oil is not new: a 2007 study found its in vivo efficacy notably similar to the commercial acaricide Acacerulen R against Psoroptes cuniculi in infested rabbits [6]. What the team says has been missing is adequate assessment of safety and efficacy specifically against canine sarcoptic mange [8]. Safety was also tested separately in 40 Sprague Dawley rats, split by body weight into four groups of 10, five males and five females each [15].
Two cautions for anyone tempted to reformulate a protocol on this. The write-up reports no negative health effects in clove oil-treated rats or dogs [16] and its headline frames the treatment as faster as well as plant-based [19], but the comparative day-14 mite counts and symptom scores that would justify "faster" are not in the summary available here.
